چکیده
A system has been developed which permits ultraviolet irradiation of samples in the probe of a 360 MHz nmr spectrometer using an excimer laser (A= 249 or 308 nm). This system is ideally suited for studying Chemically Induced Dynamic Nuclear Polarization (CIDNP) during photochemical reactions involving amino acids, proteins, and nucleic acids. The photoreagents quinoxaline and p-methoxyacetophenone prove to be the most promising for reactions witn His, Trp, and Tyr. Spin polarized spectra are presented and interpreted according the reaction mechanisms involved, the spin density distributions, and the relative g-factors of the radical intermediates. This system allows the use of a number of new photoreagents for studying protein structure and dynamics by laser CIDNP in solution.

Laser chemically induced dynamic nuclear polarization study of the reaction between photoexcited flavins and tryptophan derivatives at 360 MHz.
Chemically induced dynamic nuclear polarization (CIDNP) is generated when tryptophan (Trp), its derivatives, or Trp-containing peptides react with photoexcited flavins in a 360-MHz NMR spectrometer. In contrast to tyrosine (Tyr), we find that the nuclear polarization of Trp originates in an electron-transfer reaction. By use of a series of Trp derivatives, the unpaired spin-density distribution...
متن کاملDynamic nuclear polarization at 9T using a novel 250GHz gyrotron microwave source
In this communication, we report enhancements of nuclear spin polarization by dynamic nuclear polarization (DNP) in static and spinning solids at a magnetic field strength of 9T (250 GHz for g = 2 electrons, 380 MHz for 1H). In these experiments, 1H enhancements of up to 170 ± 50 have been observed in 1-13C-glycine dispersed in a 60:40 glycerol/water matrix at temperatures of 20 K; in addition,...
متن کاملChemically induced dynamic nuclear polarization (CIDNP) in the photoreaction of N-acetyl histidine, tyrosine, and tryptophan with 2,2′-dipyridyl and its dependence on the magnetic field
CIDNP effects for three amino acid-dye photoreaction systems (histidine, tyrosine, and tryptophan-dipyridyl in aqueous solution) are measured between 0 and 7 Tesla using a novel mechanical field cycling unit with fast digital positioning of a high-resolution NMR probe in a spatially varying magnetic field. The high spectral resolution allows to measure CIDNP patterns for different nuclear posit...
متن کامل308-nm excimer laser plus topical calcipotriol in the treatment of vitiligo; A single blind randomized clinical study
Background: There is a large variety of therapeutic agents for the treatment of vitiligo, but it still remains a challenge. Narrow-band UVB phototherapy and 308-nm excimer laser have been shown to be safe and effective for the treatment of vitiligo. Topical calcipotriol has recently been reported to enhance the efficacy of phototherapy, especially 8-methoxypsoralen plus UVA (PUVA). The goal of ...
متن کاملChemically Induced Dynamic Nuclear Polarization Studies of Guanosine in Nucleotides, Dinucleotides, and Oligonucleotides?
The nuclear magnetic resonance (NMR) technique of chemically induced dynamic nuclear polarization (CIDNP) has been used to study the reactions between photoexcited flavins and a wide variety of nucleotides, dinucleotides, and oligonucleotides. The greatest emphasis is placed on the purine nucleosides adenosine (A), inosine (I), and guanosine (G), particularly guanosine. The presence of G suppre...
